Peeling onions and then cut small pieces.


Add 200 ml of water and 2 teaspoons liquid and 2 tablespoons NACL and mix this mixture by a blender.


Put the mixture on the fire for a period of time.


filtrations for mixture.


Keep  the filtrate with alcohol like Methanol 100% in a dish.

Dissolve 5g of Giemsa power into 330ml of glycerin Mix.


Heat the solution from step 1 to 600C for 2 hour every 30min mix and shaking very well.


Leave solution at roome temperature to come cold then slowly add in 330ml of methanol to the solution from step 2 leave the solution one day.


Next day filter the solution from step 3.


The solution need to stand a period of time prior to use.


Leave for a week before use.


 


3- How to prepare Buffer in Karyotyping?2


Na2 HPO4                                   + KHPO4


    500ml Distilled water-5,95        4,55-500ml Distilled water


Why we use Buffer in this method?
